A strain of Cyclidium isolated from a sewage treatment plant in Geneva, Switzerland, was cloned and cultured under laboratory conditions before being characterized cytologically and molecularly. Information about the classical morphology and also about new molecular sequences has been obtained permitting its identification as Cyclidium glaucoma.
In the present work, we isolate another Cyclidium species, C. varibonneti, which resembles the type species C. glaucoma in morphology, e.g. similar oral structure, habitat, body shape and number of somatic kineties ( Song, 2000, Song et al., 2009). The main difference is that the former consistently has two macronuclei (vs. one in C. glaucoma ).
Cyclidium glaucoma and Cyclidium varibonneti branch first, followed by a strongly supported clade containing Protocyclidium citrullus, Ancistrum and Boveria (92% ML/1.00 BI). For the pleuronematids, the genera Pleuronema, Schizocalyptra and Histiobalantium are monophyletic and they group together with full support.
The molecular description includes the 18S rRNA sequence, and for the first time for C. glaucoma the ITS-1, 5.8S, ITS-2 sequence, and the mitochondrial Cytochrome oxidase C subunit 1 sequence. Three divergence groups were identified for C. glaucoma using a short rDNA sequence for evolutionary distance analysis.
