Onuphidae represent a polychaete family with a complex external morphology. The majority of the genera possess one or several synapomorphies, e.g. spiral branchiae in Diopatra, spinigers in median parapodia of Mooreonuphis, auricular prechaetal lobes on the anterior parapodia in combination with scoop-shaped pectinae chaetae in Nothria.

Onuphidae is monophyletic with a sister family Eunicidae. Subfamilies and genera of Onuphidae proposed by Paxton (1986a) are strongly supported by molecular data. The hypotheses on the intergeneric relationships based on morphological data are largely rejected or not resolved using the available genetic data.
